Fat May Be Linked to Memory Loss

Although problems with memory become increasingly common as people age, in some persons, memories last long time, even a life time. On the other hand, some people experience milder to substantial memory problems even at an earlier age.

Although there are several risk factors of dementia, abnormal fat metabolism has been known to pose a risk for memory and learning. People with high amounts of abdominal fat in their middle age are 3.6 times as likely to develop memory loss and dementia later in their life.

Neurological scientists at the Rush University Medical Center in collaboration with the National Institutes of Health have discovered that same protein that controls fat metabolism in the liver resides in the memory center of the brain (hippocampus) and controls memory and learning.

Results from the study funded by the Alzheimer’s Association and the National Institutes of Health were recently published in Cell Reports.

“We need to better understand how fat is connected to memory and learning so that we can develop effective approach to protect memory and learning,” said Kalipada Pahan, PhD, the Floyd A. Davis professor of neurology at Rush University Medical Center.

The liver is the body’s major fat metabolizing organ. Peroxisome proliferator-activated receptor alpha (PPARalpha) is known to control fat metabolism in the liver. Accordingly, PPARalpha is highly expressed in the liver.

“We are surprised to find high level of PPARalpha in the hippocampus of animal models,” said Pahan.

“While PPARalpha deficient mice are poor in learning and memory, injection of PPARα to the hippocampus of PPARalpha deficient mice improves learning and memory," said Pahan.

Since PPARalpha directly controls fat metabolism, people with abdominal fat levels have depleted PPARalpha in the liver and abnormal lipid metabolism. At first, these individuals lose PPARalpha from the liver and then eventually from the whole body including the brain. Theore, abdominal fat is an early indication of some kind of dementia later in life, according to Pahan.

By bone marrow chimera technique, researchers were able to create some mice having normal PPARalpha in the liver and depleted PPARalpha in the brain. These mice were poor in memory and learning. On the other hand, mice that have normal PPARalpha in the brain and depleted PPARalpha in the liver showed normal memory.

“Our study indicates that people may suffer from memory-related problems only when they lose PPARalpha in the hippocampus”, said Pahan.

CREB (cyclic AMP response element-binding protein) is called the master regulator of memory as it controls different memory-related proteins. “Our study shows that PPARalpha directly stimulates CREB and thereby increases memory-related proteins”, said Pahan.

“Further research must be conducted to see how we could potentially maintain normal PPARalpha in the brain in order to be resistant to memory loss”, said Pahan.

Other Rush researchers involved in this study include Avik Roy, PhD, research assistant professor; Malabendu Jana, PhD assistant professor; Grant Corbett, neuroscience graduate student; Shilpa Ramaswamy, instructor; and Jeffrey H. Kordower, PhD, the Jean Schweppe Armour professor of neurological sciences.

Alzheimer’s disease is the most common human disorder associated with memory loss. This disease slowly destroys memory and thinking skills, and eventually even the ability to carry out the simplest tasks. Nationwide, the total payments for services for people with Alzheimer’s and other forms of dementia will total $203 billion in 2013. By 2050, the total costs are expected to increase 500 percent to a staggering $1.2 trillion.

Musculoskeletal conditions injuries may be associated with statin use

Using cholesterol-lowering statins may be associated with musculoskeletal conditions, arthropathies (joint diseases) and injuries, according to a report published Online First by JAMA Internal Medicine, a JAMA Network publication.

While statins effectively lower cardiovascular illnesses and death, the full spectrum of statin musculoskeletal adverse events (AEs) is unknown. Statin-associated musculoskeletal AEs include a wide variety of clinical presentations, including muscle weakness, muscle cramps and tendinous (tendon) diseases, the authors write in the study background.

Ishak Mansi, M.D., of the VA North Texas Health Care System, Dallas, and colleagues utilized data from a military health care system to determine whether statins were associated with musculoskeletal conditions based on statin use during the 2005 fiscal year. Patients were divided into two groups: statin users for at least 90 days and nonusers. A total of 46,249 patients met the study criteria and of those, researchers propensity score-matched (a statistical approach that mathematically matches the characteristics of patients in two or more groups) 6,967 statin users with 6,967 nonusers.

"Musculoskeletal conditions, arthropathies, injuries and pain are more common among statin users than among similar nonusers. The full spectrum of statins musculoskeletal adverse events may not be fully explored, and further studies are warranted, especially in physically active individuals," the authors notes.

Statin users had a higher odds ratio (OR) for musculoskeletal disease diagnosis group 1 (all musculoskeletal diseases: OR, 1.19), for musculoskeletal disease diagnosis group 1b (dislocation/strain/sprain: OR, 1.13) and for musculoskeletal diagnosis group 2 (musculoskeletal pain: OR, 1.09), but not for musculoskeletal disease diagnosis group 1a (osteoarthritis/arthropathy: OR,1.07), according to study results for the propensity score-matched pairs.

To our knowledge, this is the first study, using propensity score matching, to show that statin use is associated with an increased likelihood of diagnoses of musculoskeletal conditions, arthropathies and injuries. In our primary analysis, we did not find a statistically significant association between statin use and arthropathy; however, this association was statistically significant in all other analyses," the authors conclude. "These findings are concerning because starting statin therapy at a young age for primary prevention of cardiovascular diseases has been widely advocated."

A British scientist has become the first human to be infected with a computer virus.
Dr Mark Gasson, a cybernetics expert at the University of Reading, has had a computer chip implanted in his hand.
The chip is programmed to open security doors to his lab - and ensure only he is able to switch on and use his mobile phone.
But Dr Gasson deliberately infected the chip with a computer virus, which was then automatically transmitted to the lab security system.
"Once the system is infected, anybody accessing the building with their passcard would be infected too," he told Sky News.
The virus on his chip is benign. But malicious computer code could give criminals access to a building.
Dr Gasson says his experiment also exposes the vulnerability of chips now routinely implanted in patients.

Heart pacemakers contain mini-computers that control the heartbeat, and communicate with doctors via a special reader held against the skin.
But if a virus was transmitted to the device which stopped it working properly, the consequences for the patient could be fatal.
"The devices will have to start to use security encryption," said Dr Gasson.
"Medical devices should have some kind of password protection as well. Theyre basic security precautions. Its surprising these devices dont have them already."

Resveratrol can still be used in the body after it has been metabolized

A chemical found in red wine remains effective at fighting cancer even after the bodys metabolism has converted it into other compounds.

This is an important finding in a new paper published in the journal Science Translational Medicine by Cancer Research UK-funded researchers at the University of Leicesters Department of Cancer Studies and Molecular Medicine.

The paper reveals that resveratrol – a compound extracted from the skins of red grapes – is not rendered ineffective once it is metabolised by the body.

This is an important development, as resveratrol is metabolised very quickly – and it had previously been thought that levels of the extracted chemical drop too quickly to make it usable in clinical trials.

The new research shows that the chemical can still be taken into cells after it has been metabolised into resveratrol sulfates.

Enzymes within cells are then able to break it down into resveratrol again – meaning that levels of resveratrol in the cells are higher than was previously thought.

In fact, the results appear to show resveratrol may be more effective once it has been generated from resveratrol sulfate than it is if it has never been metabolised because the concentrations achieved are higher.

The team, led by University of Leicester translational cancer research expert Professor Karen Brown, administered resveratrol sulfate to mice models.

They were subsequently able to detect free resveratrol in plasma and a variety of tissues in the mice.

This is the first direct sign that resveratrol can be formed from resveratrol sulfate in live animals, and the researchers think it may help to show how resveratrol is able to have beneficial effects in animals.

The study also showed that resveratrol generated from resveratrol sulfate is able to slow the growth of cancer cells by causing them to digest their own internal constituents and stopping them from dividing.

Professor Karen Brown said: "There is a lot of strong evidence from laboratory models that resveratrol can do a whole host of beneficial things – from protecting against a variety of cancers and heart disease to extending lifespan.

"It has been known for many years that resveratrol is rapidly converted to sulfate and glucuronide metabolites in humans and animals – meaning the plasma concentrations of resveratrol itself quickly become very low after administration.

"It has always been difficult to understand how resveratrol is able to have activity in animal models when the concentrations present are so low, and it has made some people skeptical about whether it might have any effects in humans.

"Researchers have hypothesized for a long time that resveratrol might be regenerated from its major metabolites in whole animals but it has never been proven.

"Our study was the first to show that resveratrol can be regenerated from sulfate metabolites in cells and that this resveratrol can then have biological activity that could be useful in a wide variety of diseases in humans.

"Importantly, we did all our work with clinically achievable concentrations so we are hopeful that our findings will translate to humans.

"Overall, I think our findings are very encouraging for all types of medical research on resveratrol. They help to justify future clinical trials where, previously, it may have been difficult to argue that resveratrol can be useful in humans because of the low detectable concentrations.

"There is considerable commercial interest in developing new forms of resveratrol that can resist or overcome the issue of rapid metabolism. Our results suggest such products may not actually be necessary to deliver biologically active doses of resveratrol to people."
